BUT PROFOUND NAIVETÉ LED ME TO THINK “HOW COULD I NOT SUCCEED.” WE THEN SET ABOUT REVOLUTIONIZING AMINO ACID ANALYSIS. Previously you could do amino acid analysis only with a dedicated amino acid analyzer which has almost no fl exibility. We developed an eluant/column combination and the hardware necessary to transfer the analysis to a modern gradient HPLC. The result was a method with the most fl exibility that could evolve with the hardware and thus benefi t from each generational improvement. Thus the user may always have the benefi ts of the latest HPLC technology. Even today, dedicated amino acid analyzers execute the same method as when introduced in the late 50’s. MICHAEL V. PICKERING, PH.D. / FOUNDER As we enter a new decade, I am drawn back to the beginning. After I finished two postdoctoral positions in medicinal chemistry, I worked four years in R&D at two different HPLC instrument companies. During that time, I came 01 to the realization that the end user does not want capital equipment, solvents, chemicals, or computers. They want reliable chromatograms. As Aristotle’s theory on causality teaches, the final cause comes first. In this instance, the final chromatogram drives the user’s choice for equipment and chemistry. So convinced was I of this truth, I jumped into the void of self- employment. Our product would be the ethereal “Guaranteed Chromatography.” I started Pickering Laboratories in 1980, with the patented ninhydrin reagent TRIONE®. Soon thereafter we began offering gradient Amino Acid Analysis methods that were achievable with modern HPLC equipment. Back then, Amino Acid Analysis run times were 140-minutes for Lithium cation-exchange (native fluid analysis) and 90-minutes for Sodium cation-exchange (hydrolysate analysis). Today, the same chromatograms are achievable in 70-minutes for Lithium and 30-minutes for Sodium. As you are all aware by now, we work hand in hand with the AOAC, FDA, USDA and USEPA. The relationships we’ve cultivated over the years with these regulatory agencies have enabled us to influence analytical methods as they develop, and to better understand and support you. In the same context, we are regularly invited to make custom formulations and develop pharmaceutical QC methods. Our contribution to these regulatory agencies was recently recognized in September of 2009, at the 125th Anniver- sary meeting of the AOAC International in Philadelphia. Pickering Laboratories received AOAC’s Single Labora- tory Validation of the Year Award for Multiresidue Mycotoxin Analysis in Corn Grain by High-Performance Liquid Chromatography with Post-column Photochemical and Chemical Derivatization. Several colleagues and I attended the award ceremony – it was an honor for Pickering Laboratories. In fact, the award was doubled to include our Single-Laboratory Validation for the Quantification of Neomycin B and Neomycin C in Animal Feeds by Liquid Chromatography Fluorescence Detection with Post-column Derivatization. Well, here we are, 30 years later. My belief in Aristotle’s theory has turned out to be true. The relationship we have created has knit us together as a team: you observe/we support. This feature is useful in Amino Acid Analysis by enabling additional fl exibility when optimizing methods. Using the temperature gradient program allows for improved separation, shortened analysis time and fi ne-tuned methods for specifi c compound detection. The Chromatogram in Figure 1 demonstrates some of the clear advantages of the temperature gradient program. Com- pared to the standard method without the temperature gradient the total run time is 42 % shorter. Additionally, separation is improved.
